This will come through natural play. Personally I unlocked it before finishing Sonic 1 whilst in story mode

It may not unlock as the 50th enemy is destroyed, but rather when the act is cleared after defeating at least 50

Just play whatever game and jump on enemies' heads to kill them.

Enemies are plentiful throughout all Sonic games and you defeat many of them as you progress through the games, so this will also come naturally as you play.
