Each time you beat one of the bosses in your first playthrough of the game (there are three), you earn a Space Jam card. The remaining two cards to be earned are acquired by completing Boss Rush Mode for one and beating the game on Hard difficulty for another.

The guide is right about the cards from the three bosses, but as an aside, you earn the last two cards by beating Boss Rush (gives you Daffy Duck) and Hard Mode (gives you Marvin The Martian).

Space Jam Cards are power ups that can give you increased health or speed. You will start the game with a couple already unlocked. The rest are unlocked by defeating each boss in the main game (one card per boss). Then, after completing the main game you will unlock Hardcore mode and Boss Rush mode. Completing both of those will reward you with your final two cards.
