This one's a grind.
In order to unlock the dungeon levels, you must first complete all 100 levels in the tower. Then you can access your current dungeon level by pressing
. There is no level select with in the dungeon- you only have access to the current level.
The dungeon is structured differently than the tower. Levels are endless and you play until you die. On each level, you are given three tasks to complete. Complete all three to earn a trophy and get three new tasks. You must earn five trophies to advance to the next level.
Tasks requirements are things like "kill 4 enemies with palindromes," "kill 12 enemies in one run," "find 12,000 gold," etc. Progress is carried across each run unless noted otherwise on the task (so you could kill one enemy with a palindrome, then die, then repeat three times for example).
Even with an anagram solver to maximize efficiency, it takes about 20 minutes to advance through a single level. So you're looking at at least 15 hours doing the same tedious stuff in the dungeon to get to level 42 for this achievement.

Just one thing to mention here. You can restart the level as many times as you want and your progress will carry over as long as you die in the end.
So if, for example, you need 4 palindromes, you can get two, restart until you have the necessary letters, get two more, and then die to "cash in" what you have done.
Quitting the level without dying, however, will result in you losing all progress you have made since your last death.
