This is best done on act 3 after you connect all the DNA carts just keep using your X and Y combos the big thing is to use your left trigger decoy ability Here is a video to help you get this done.
If you run out of enemy that are close up the web shot will keep your combo going but will not add to your combo.

I wound up getting 197 and losing my multiplier LOL

I would just like to add two things:
1.) Once the the percentage in the corner gets to 85% more enemies will spawn and freeze everyone just long enough to break your combo. If you find yourself not at 200 hits and the time running out, charge your spin kick move. If you time it right you can continue hitting enemies and building your combo as the enemies appear.
2.) The distance grab (rb enemies far away) was a life saver here. Pulling enemies that did not want to cooperate with me getting the cheevo made this is as easy as can be.

The best place I found was the spot after you come back up to the DNA lab. When you are swarmed by all the failed experiments and you have a bunch around you, use pain tornado as much as possible, and don't worry about messing up cause if the next part starts and amazing spider-man takes over, just quit. Go back to the main menu; this should put you back at the beginning of the fight. This might be hard, it took me a few tries but I got it using this method. It also helps to put it on “easy”, that way you don't get killed so easily. In the words of Stan the man “good luck true believers”.

It's the right place to do so. It's even easier if you have Roundhouse Spin. Keep pressing X if you have it and the achievement will come. I got it on my first try.
