Playing Doctor and Heal Thyself can be done together though Heal Thyself will be unlocked first simply because you need to cure yourself at points where you must.
You will need 100 Cases of Meds and 300 Blood Plague Samples to craft 100 Plague cures, you may reasonably need to craft more just in case you used it on yourself otherwise you're in the process of doing it earlier than going through this guide.
Easiest way I did this was having a follower follow me and both carry cures after farming for the materials. Then walk into the wall and switching to my follower and tell them to use the cure.
You basically have them both walk into the wall and out to gain plague and tell each other to use the cure than on yourself. Two birds, one stone.
Build an outpost nearby just in case

If you don't want to duplicate plague cures and need to farm them yourself, this is an addition to this guide:
You're going to need a ton of plague cure for this so it's important to have an efficient way of getting it.
- Farm an epic ass-ton of Plague Buster (150-200+). Use your radio to find Bloater locations on cooldown. To speed the cooldown up, fill up all of your outpost slots with 3-4 of the Signal Booster outposts. These decrease the cooldown, and I'm pretty sure they stack. If you're doing a bunch of radio calls in a row, it was very common that the Bloaters would respawn right next to me at the same area I was just at. This really cut down the time it took to accumulate tons of Plague Buster.
- Create a bunch of Fireworks.
- Go to a Plague Wall. Your Character should be Isaac. Your follower can be anyone.
- Inventory: Have a high capacity magazine gun (100 rounds or more), a few pain killers, and both of your characters should have inventories full of Plague Buster and Firecrackers.
- Throw one Fire Cracker, then throw one Plague Buster at the wall. Do not kill the wall.
- Assuming you do this on the Fourth Wall (the last one) the first segment wave will spawn tons of zombies and always spawn a Juggernaut every time you throw a Plague Buster at the wall and make the Wall vulnerable to damage. The Juggernaut will drop 2-4 Samples every time you kill it, and everything else is just bonus. Killing the Juggernaut quickly by using 2 Plague Busters then melee executing it will make you have to burn through more mats, but this is the fastest way.
- Rinse repeat until out of Plague Buster. You'll want at least 3 Plague Buster and one Fire Cracker per wave. Firecrackers only cost 10 parts to make. The limiting thing here will be Plague Buster, which isn't that hard to get.
Using this method, in about 15 minutes I can get 80-100 Plague Samples. This obviously doesn't take into account the time to farm Plague Buster.
To get meds, use your radio and make meds in your infirmary on cooldown until out of Chemicals. After you're out of chemicals you can just use your radio, and optionally convert as many outposts as you want to into medical outposts. I'd farm your Bloater Poison Glands (using 3-4 signal booster outposts mentioned above) first since you can hold as many of those as you want, then farm meds afterwards and convert the outposts into medical outposts as you're farming them.
