Will the infestations respawn or do you have to restart the game?

Just for clarification I can confirm that 'one day' is any 24 hour period in real time. I did infestations yesterday and today and unlocked it.

i have done 5 within 20 minutes and no achievement same problem with the kill 10 horde in a day

I did five within 24 hours, too. No achievement. I know it's accurate because I'm tracking the Challenge and I'm at 40%. Maybe it is a 24 hour in game time?

A strategic and easy way to get this achievement in one shot that preserves your vehicle(s) and doesn't require you to go home to resupply or heal between infestations:
Once you get five infestations on the map, hopefully at least in the same general area, select a person with the shooting skill and focus aim special technique, and give them the 9mm pistol with silencer attached, (one that holds at least 10 rounds - the 17 round 9mm is the best I've found). Also bring extra food, 9mm ammo, and meds. Get one of the big passenger trucks, and pack some meds, food, and bullets into the truck just in case you have any problems.
Drive the truck out to an infestation, stop outside the house and just keep hitting the horn, this will get most of the zombies out, and usually the screamers as well. Now keep backing over the zombies. If you back over them, it does zero damage to the truck, so you can keep backing over them, and any hoards you will probably attract. You can also hit them with your door, just don't drive into them head on. In Year One you can get a ton of zombies on the screen at once, so backing over the mob of zombies you attract, plus the extra hoards that might be in the area is really the only realistic way to survive in extreme cases When the street is clear, head inside, take out the rest of the zombies and/or screamers, and clear it. Do that five times.
If you find yourself stuck outside the truck, that's where the focus aim comes in real handy - take out as many zombies as you can quickly, and jump back in the truck. Obviously the food helps out so you can maintain aim, and meds if you get too damaged. You can clean out all five sites in less than 10 minutes this way (depending on driving distance), without wrecking a vehicle or being forced to return home, and it's pretty safe for your character as well.
