This mission unlocks with floor 22 in the DLC. You can back out to the main mission screen to find it. You can play it at power level 300 and with extra mode+Runic Protection for another easy boss fight.
Edit: My game gave me the mission on floor 20, but a friend didn't get it until 22. I've updated the guide to say 22, but it may unlock sooner.

I've gotten to floor 24 and I still don't have the mission. It seems random.

"Chaos Rift - Battle on the Big Bridge" is a side mission that will be added after some progression through the Rift Labyrinth and which can only be played on GILGAMESH difficulty.
You will face again the recurrent boss of the Labyrinth. Fortunately, you can rely again on Extra Mode + Liberator, as you will not have any problem of max HP reduction.
Once you complete the battle, this trophy will pop.
