To unlock this achievement you must simply complete the story mode in single player on any difficulty (preferable on easy) as Estel without changing characters.

Does it have to be in 1 sitting? I just beat Story with her on save file 1 (3rd time continuing story because this game honestly annoys me, so did a few levels per night). No pop.

You shouldn't have to do it it in one sitting, I looked at my achievements and I got these types of achievements within multiple sittings (Unless they updated it to where you need to or DLC characters work differently). The other thing I remember is that the achievement doesn't unlock after beating the final boss but at the results screen after the credits.

This trophy is self-explanatory, you need to finish the 12 stages of Story mode with Estel, a brand new Streets of Rage 4 character which should be available as soon as you install the DLC. It can be done on Easy difficulty and using Chapter Select from the main menu. Please note that despite being added as a playable character with this add-on, everything done with Estel in the main game also increases your Lifetime Score.
Carmona, Caramel, Cardinal
Clear all 12 stages with Estel in single player story mode. Do not switch characters and be sure to watch the stage summary at the end of the final level or the achievement will not unlock, This can be completed on Easy difficulty though.
