This achievement is not as hard as it sounds.
Getting a 50x combo on the boss itself would be difficult. Fortunately, you only need a 50x combo in the stage, not necessarily against the boss itself.
First, you will want to have purchased the perk for getting x2 hits. Equip it in the 30 combo slot. Once you get a combo of 30, each regular attack will give you two hits (but more importantly, two points on your combo).
Once you knock off a fraction of the boss' life bar, he will run away and some enemies will spawn. The first wave will be common enemies. Just spam cn_X on these guys and, with the x2 hit perk, you should have over 50x combo before you clear the wave.
