There are two ways to accomplish this 'heroic' ending using no continues (aside from destroying all the hydorah seeds before beating the final level)
1) Simply be familiar with all the levels, and good enough to power through the game on a single playthrough (there are a number of extra lives you can grab along the way, so you can die some), or;
2) Before losing your last life on a given stage, simply quit out of the game and then reload. You can start back up at your most recent save (should be the last level you beat).
On my first playthough, I used method two. Levels with hydorah seeds are:
Dunarys
Path of Scylla
Underground Base
Black Gate*
*You must defeat the Rubina level and receive the Light special 'weapon' to access the final seed at Black Gate. Activate the Light and use it to light the torches to open up the path at castle on the surface and then later to pull the final seed out of the pool so you can destroy it.
