I want to start by saying that you can get every achievements with the Assist ON (Bottom left corner in Play Game)
If you finish a playthrough and you still need this achievement or the 1000 enemies killed, you can start a new game and your progress will carry over. The 250 diamonds achievement unlock at my 2nd playthrough in the 3rd area.

This was the final achievement I had left to mop up, and my life was made a little easier on Stage 3 - The Atmosphere.
About half way thorough the stage, you enter a bit of tunnel structure where some large silver ships attack you. If you destroy them, they kick out 5 diamonds each. There are quite a few of these ships in this section, so you can easily accrue 20 to 40 diamonds (if not more), per 'run'.
If you are reading this before your first play-through, then I’d recommend rinsing this section a few times when you first get to it - you get an extra life when you enter the tunnel, so destroying all the ships and purposely dying a few times shouldn’t really affect your life count.
If not, then getting back to this section on stage 3, from a new game, doesn’t take too much time once you’ve picked up the rest of the achievements…

A good way to grind this out is by using Custom Mode to start at level 3.
Custom mode is unlocked by beating the full game once - even with assists enabled. You'll likely not have enough diamonds by the time you get the last of the boss achievements, and the final two levels are fairly easy.
Once custom mode is unlocked, start a game with speed maxed, shield at 9000, lives at 99 and starting from level 3. I also recommend using the scatter shot.
Run through level 3 collecting every diamond/item you can (as another solution says, the big silver guys drop 5 diamonds each, use this) and restart if you get to the corridor with downward shooting guns. Only costs 1 life. Repeat until unlocked.

This achievement is cumulative across all levels and playthroughs.
The achievement does not depent on color of the diamonds. Diamonds drop quite often as you play, with some of the larger enemies dropping a bunch of diamonds. If you make it a point to collect most of the diamonds you see, you'll likely unlock this achievement by the end of your first playthrough. Since diamonds are so plentiful, don't put yourself in harm's way just to collect one or two diamonds. It's easy and fast enough to simply start a second playthrough and grab all the diamonds you see in the first level to mop up whatever you need for this achievement.
