Defeated Duke using a mystic arte.

How to unlock the Secret Mission 23 achievement in Tales of Vesperia - Definitive Guide

You will need to defeat the final boss of the main storyline, Duke, in his second form, using a Mystic Arte.

To use a Mystic Arte, you must have the "Special" skill activated and equipped, as well as having overlimit 4 synthesized.

When fighting Duke, use a Magic Lens on him and then keep an eye on his HP remaining using RB. When he gets into the 25,000 range, put your characters into a full defend strategy. If Duke currently has an overlimit active, wait for it to expire, as he will likely use his own Mystic Arte and interrupt you otherwise. When ready, active overlimit 4 by pressing up on the D-pad, use an arcane arte, making sure to hold down B button. Your Mystic Arte will trigger and should deliver the fatal blow on the boss.

Depending on the character you are using, and your level relative to Duke's, your mystic arte may not do enough damage to kill Duke (Estelle's for example has a healing component so is not as damaging as Yuri's). If you don't happen to finish him off, you can reload your save or use enough limit bottles to trigger another mystic arte and try again.

Instead of making a new solution, I'll just comment.

I was lvl 60 using Yuri as my controlled character, Estelle, Karol and Rita were the NPCs. Once Duke got below 30,000 I put Karol and Rita on defend only allowing them to only use items. Kept Estelle on Heal only. Once Duke was at 15,000 I used my overlimit (NO BURST, JUST MYSTIC). I ended up inflicting around 22,000 to him.

If your at a higher level, it'll be easier, just do this around 25,000 - 30,000, lower at around 10,000. Your Mystic arte gets a little extra move at the end when fighting Duke, it's bad ass.

At lvl 200 this is pretty tricky if you have your fell arms pretty high up there, on your 2nd playthrough. I had to do it around 50K to trigger the arcane arte, as it does about 20k dmg with that alone -_- Then I was able to use my Mystic Arte, which ended him.

You're going to need Duke into his second form (the white one), then get him down to about 20k for this to work. It could be a little more, especially if you're on easy difficulty, but just to be safe go low. Once you get there, put your allies on defend and go into Overlimit (make sure it is level 3 or 4) and use an altered or arcane arte, then hold the B button while you are performing the arte. If done correctly, you will launch into your Mystic arte and should do enough damage to kill him.
