This can only be done in Arcade Mode.
You will need at least two controllers to unlock this achievement.
The original achievement description states that you need to get the most melee kills in a multiplayer match, but the achievement will actually unlock in the middle of the stage after getting 2 or 3 melee kills with the character you selected under your main account.

Just a comment that I was able to unlock this today by playing a Story Mode level with two contollers as well
