This is cumulative across all game modes and episode replays. Each episode seems to always have two or three set pizzas along the way, and one pizza right before the boss that fully heals all party members. You'll therefore be getting a solid three pizzas per episode, and considering there are 16 episodes, you'll unlock this achievement before finishing Story Mode.

Pizzas are hidden throughout each level of the game, typically under breakable objects, and will refill your health. They can only be picked up if you've taken damage, and in multiplayer, typically only the character that picks the pizza up will refill their health. You will certainly unlock this over the course of your playthroughs.
