Going for the no damage achievements is actually very easy using an exploit. Playing as the Lost is exactly like this since he can't be damaged at all without dying.
Everytime you back out to the main menu you get an autosave. So keep that in mind.
After you've cleared one or more (how many you like) rooms without being damaged exit to the main menu to get another autosave.
In case you get hit you have to either let enemies, spikes or bonfires kill you or bomb yourself to death. This can be a tad annoying if you have 12 hearts towards the last couple of floors like the "Womb/Utero" or the Darkroom/Chest.

NOW THE IMPORTANT PART!!!
During the death animation pause the game and hit the guide button. Manually quit the game.

VERY IMPORTANT!!!
You MUSTN'T quit after being hit BEFORE you die. That way it would for whatever reason save your current health.
VERY IMPORTANT!!!

If you continue after starting the game up again you will be back where your last autosave was.

This works for THE LOST playthroughs as well!!!
Of course with him killing yourself is easier since he has no health and dies in one hit.
Items like Holy Mantle (you can take one hit every room), the "9 lives cat head" or guppy's collar (1 extra life) can change this, though.
Avoid the Ankh at all cost since it respawns you as another character and would therefor void the achievement. Same goes for Judas' Shadow and Lazarus' Rags.

So just pray to the RNG God to be lucky with drops and this is going to be a breeze. Time consuming but easy.

I'll try this soon but I guess that if you posted it that means it works so congrats for finding this. :]

I hope they won't patch it. Anyway i won't update my game until they fix some achievements.

I haven't really played with the Lost yet since I wanna do everything else first. But I tried for a bit if you can can the save scumming with him as well and can confirm that it does work indeed.
I'm gonna hold of with beating the Chest and Dark Room with one character since those are glitched and I don't know how the achievement is gonna be triggered when it's gonna be patched.
Same goes for the 20 challenges. I'm gonna leave an easy one unfinished until we get a patch.

Yup I just tried myself and this works.
To be honest I'll just use this glitch for The Lost runs otherwise it will ruin the fun of the game.

According to PS3-4 users who got the game way before us and had achievements problems too, once they released some patches their trophies unlocked automatically but yeah we can't be sure it'll be the same for us.

I only used this glitch for the harder no damage runs and plan on using it for the Lost since this character sucks balls.
It's only gonna be endless restarting until holy mantle shows up in the first item room and then some luck with the RNG.
Rest should be a cakewalk using this method.

Can you use a seed to unlock the achievement. My brother found a seed online that had an easy way to get to Satan and Lucifer. He killed both of them and the achievement never popped. Any reasons why?

You can only use seeds for unlocking characters. There's a good solution for the "unlock the lost" achievement.
But seeds disable any other achievement progress...

I believe the Afterbirth expansion has fixed the save exploit.

Unfortunately he's right, I've just tried to use the exploit and it doesn't seem to work with Afterbirth installed.
On the plus side The Lost starts with Holy Mantle after you've donated 879 coins to the Greed Machine and Afterbirth adds a whole load more items including a bunch of very powerful synergies which can be used to make the game MUCH easier.
That being said you may still be better off getting your 6 red markers with The Lost before buying Afterbirth, that save exploit is a lifesaver.

I think I found an exploit that seems to be working in Afterbirth!

If you die, disconnect from the internet, force quit the game, and then reconnect to the internet and start the game up again, you'll be in the same room with 0 health... and will die again. Sounds useless.

HOWEVER, if you have Guppy's Collar and you die because it didn't proc, YOU CAN USE THE ABOVE STEPS UNTIL IT DOES.

Requires finding Guppy's Collar but holy hell it's a good exploit.
