Cards are earned after a mission and are typically either level 1 or level 2. To get a card to level 3, which is their maximum before any upgrades with Madame Mei, you have two choices:

• While highlighting the card, press (F on keyboard) to expend Ink to level the card.

• Assign an identical card to the same slot.

Ink is gained from discarding cards (you can level up how much Ink you get with Madame Mei) and only used to level up cards. Upgrading a card from level 1 to level 2 costs 32 Ink. The cost increases to 128 Ink when going from level 2 to level 3.

I would recommend keeping your Ink until you draw a card that you already have assigned and want to upgrade. You can then choose to first upgrade to level 2 before you assign the duplicate card which will bump the level to 3 and get you the achievement.
