You will fight Manfredd the Accursed right after you fight Lisa in the Ordens Regnant Palace. After you fight Lisa, make sure to activate the checkpoint in the previous room. After beating Manfredd you will get the corrupted heart of Nycrux and in the next room you will finally get the Celestial Larva aka double jump.
