Story related and cannot be missed.

When starting the game, follow the yellow star and follow the instructions to eventually build a micro-manager that will break the door and unlock Lucy.

This is story related and cannot be missed. This unlocks in Chapter 1, 'Apocalypeburg'. Follow Unikitty's instructions and walk over to the Apocalypse Dome. There you will see an imprisoned Lucy. Smash three objects and collect the LEGO pieces to build the Micro Manager which will release Lucy.
