The Highway (Also called Durin's Highway) can only be accessed by first crafting Durin's Axe. The entrances to the highway can be identified by large rock doorways covered in purple smoke. After crafting the axe, you can run up to the doorways and break them down, revealing the highway beyond.
Finding the different fragments of Durin's Axe can be tricky, as the locations seem to have moved around slightly between playthroughs. I know for a fact the first fragment is found BEFORE entering the Elven Quarter (so if you didn't find one, go check the area). And another fragment can be found after defeating Bolgakh in the Mines of Moria.
There is an achievement for crafting Durin's Axe, so hopefully someone will have commented here or there on where they found their fragments and we can all help each other!
