Non-spoiler solution:
The achievement is story related and cannot be missed. It pops during one of two mutually exclusive main story missions. During the main story mission Naked Hunch, you must side with one of two people. This branches the game towards a good ending and a bad ending. Depending on your branch, you will encounter the slug queen in either The Beast That Shouted Love at the Heart of the World or For a Breath I Tarry. You must kill her to advance the story unlocking the achievement.

Either branch allows you to complete the DLC and get all the achievements. So this achievement is unmissable although the story is significantly different depending on your path. For this reason, you may want to make a manual save during Naked Hunch either just before or just after going up the distillery elevator. If playing on supernova, you'll have to make the manual save on your ship before going to the distillery. You can then revert to this save for the other ending noting that it would lead to no additional achievements.

Spoiler solution:

Once you investigate any suspect and report back to Ludovico, you make the main story quest An Unfinished Accusation available. If you want the Interrogo Totus achievement, you will want to investigate all suspects and report back to Ludovico but you can advance the story with only one.

In the dialog associated with An Unfinished Accusation, name any suspect as the murderer. The choice does not matter. Ludovico is the murderer but you will have to pass a high engineering skill check (around 105) to prove it. Since it does not matter whom you name, don't worry if you don't have a high engineering skill. Name whomsoever you want.

Naming a suspect with any of the 5 choices—the correct one (whether or not you can pass the skill check) or any of the 4 incorrect ones—leads to the same outcome. The distillery is attacked and you have to go rescue Ludovico. This concludes An Unfinished Accusation and initiates the main quest, Naked Hunch

Make sure Naked Hunch is active. This is the point where you will want to make a manual save if you want to see both endings—on your ship if playing on supernova or before entering the distillery if playing on any other difficulty mode.

Follow the quest marker to the distillery—you take the bridge on the right front sector of the hotel island as shown on your map. Follow the quest marker to the left side of the building to find an elevator. Take it. You'll see a switch with a button prompt.

This is the branch point for the two endings. If you are not playing on supernova, this is your last chance to make a manual save if you are interested in seeing both mutually exclusive endings. It must be made before pressing the button prompt. Nevertheless, for the sake of a 100% achievement completion you only need one ending so making the save to see the other ending is solely for your personal edification.

Pressing the button reveals Helen on the other side of a glass plate. Ludovico did not murder her but her twin sister Belinda. In the ensuing dialog, you must pick a side. If you side with Helen, you activate The Bird Drug mission. If you side with Ludovico, you activate The Story of Two Disasters main story mission. Either one allows you to kill the slug queen for this achievement and complete the DLC for the Murder on Eridanos achievement. Siding with Ludovico for the bad ending is a little simpler if you're into expediency.

Siding with Ludovico: The Story of Two Disasters transitions into the main story mission, For a Breath I Tarry where you encounter the slug queen. Siding with Helen: The Bird Drug transitions into the main story mission, The Beast That Shouted Love at the Heart of the World where you encounter the slug queen. Though the circumstances leading up to the encounter are significantly different, the result is the same. You must battle and kill the slug queen. Doing so unlocks the achievement.

Note regarding Spectrum Needler Buddy achievement: If you side with Helen, persuade her to give you the Needler gun for the Spectrum Needler Buddy achievement when you meet up with her in the penthouse. If you side with Ludovico, you will encounter Helen at the end of The Story of Two Disasters. When you do, kill her and loot her body to get the Needler. You also have the option to pickpocket her for the weapon.

Absolutely. I've been playing stealth primarily. My go-to weapon for Murder was the single shot Dead-Eye Assault Rifle III introduced in this DLC. As a single shot weapon, its two shots per second doesn't have the rate of fire of a light machine gun but each round inflicts 459 damage at level 41. The mag-to-melt mod is particularly lethal against the enemies in this DLC. More importantly, it's got the execute special effect. On Supernova, it takes about 6 rounds to kill the Queen--one magazine--and can do so before tactical time dilation runs out.
