No spoiler solution: This is story related and cannot be missed. During the main story mission, The Naked Hunch, you must pick a side leading to one of two main story missions: The Story of Two Disasters or The Bird Drug. Completing either one will unlock the achievement.
Spoiler solution:
Once you investigate any suspect and report back to Ludovico, you make the main story quest An Unfinished Accusation available. If you want the Interrogo Totus achievement, you will want to investigate all suspects and report back to Ludovico but you can advance the story with only one.
In the dialog associated with An Unfinished Accusation, name Ludovico as the murderer. This precipitates an attack on the distillery. Go there to rescue Ludovico--it's on the small island to the northeast. The mission automatically transitions from An Unfinished Accusation to Naked Hunch. Follow the quest marker to the left side of the building to find an elevator. Take it. You'll see a switch with a button prompt.
Before pressing the prompt make a manual save if you are interested in seeing both mutually exclusive endings. If you are playing on supernova, make the manual save on your ship before going to the distillery. For the sake of a 100% achievement completion, you only need one ending. So, the save is completely at your prerogative if you desire to see both endings.
If you are going to be continuing the game to the end of the story, note that the endings have a critical impact on the repercussions revealed after the finish. The The Bird Drug is the good Eridanos ending. Endings have no impact on achievements. Nevertheless, if, for personal reasons, you are pursuing the game's best ending make sure The Bird Drug path is the one you pursue and carry forward.
Pressing the button reveals Helen across from a glass plate. Ludovico did not murder her but her twin sister Belinda. In the ensuing dialog, you must pick a side. If you side with Helen, you activate The Bird Drug mission. If you side with Ludovico, you activate The Story of Two Disasters main story mission. Either one allows you to kill the queen for the You Have Selected Regicide! achievement and complete the questline for the Murder on Eridanos achievement.
Completing either The Story of Two Disasters for the 'bad' ending (infect Eridanos) or The Bird Drug for the 'good' ending (cure Eridanos) unlocks the achievement. Siding with Ludovico for the bad ending is a little simpler if you're into expediency. As stated earlier, you can revert to a manual save made before talking to Helen. Making the opposite choice of whom to side with will lead to the other ending.
Note regarding Spectrum Needler Buddy achievement: If you side with Helen, persuade her to give you the Needler gun for this achievement. If you side with Ludovico, you will encounter Helen later. When you do, either pickpocket her or kill her and loot her body to get the Needler.
