You need to set up a local game vs. the AI: Main menu - Local - Single Player. Set the board size to 9, difficulty to 3 or higher, and the other settings at default.

So what's the difference between these two? Can I unlock them at the same time?

Yes, they can be unlocked in one game, if you play the AI on the highest difficulty. This achievement requires AI level 3 or higher, Better than silicon requires level 5. The scale is 1 (weak) to 5 (strongest).
