Unfortunately this must be 100 different animals. You can't just find 2 deer standing together and keep marking them back and fourth. I am only assuming this doesn't work for 100 as it doesn't work for 10.

To mark an animal, you must pull out your binoculars and press X on the animal to mark. Use the D-Pad to zoom if the animal is too far away. This is useful to see what type of animal you are looking at but honestly I find it completely useless. The lack of animals in the game and the time an animal stays marked makes marking animals quite annoying.
