Story related and cannot be missed, you will get this after completing Chapter 3: Dirty Secrets.
WARNING: There is a glitch in this mission, sometimes a rope you need to climb at the end will not appear. This can be countered by buying rope arrows before the mission, these can be used to bypass the glitch. Make sure you have rope arrows and don't lose an hour of progress like I did!

Thanks for posting the solution but I gotta tell ya; buddy who made it got really lost on the last part with the bridges and the towers. The correct path is fairly simple. You only need to turn each of the three bridges once. Bridge I (they are marked) goes clockwise and you cross it right away. You turn bridge II counter-clock-wise and you can't cross it, so you go around. Bridge III gets turned clock-wise, and you cross and then drop down to II. Eventually there is a bridge IV but it does not rotate.
