A major is any tournament rated four stars or higher. Simply win the same tournament in consecutive years for the achievement.
The earliest way to get this is to win the Masters twice in a row, which will happen if you're going for the Unstoppable! achievement.

Upon winning the Master as an Amateur, the following years Masters tournament will be your first opportunity to defend a Major title. Simply win the Masters the second year and this will unlock once the tournament is over.

Your first opportunity to defend a Major will be your first Masters as a pro, returning after winning it as an amateur. Just make sure to get your world ranking high enough to qualify for the competition as it comes around again.
If you are having problems, drop the gameplay difficulty to Amateur.
