This can be done legit against the CPU or with another controller assigned as a second player who's idle.

Simply run into any of the corners of the ring and press B to climb up onto the turn-buckle. Then press either X or A to do an aerial move onto your opponent - it must connect and do damage to them, if they counter it does not count. You will know when it has been done correctly as you will see a + with a number under your HUD display.

This can be done with any character and in any mode.

Many types of moves are counted as "aerial" moves. This includes running attacks, RT to run, and while running press X for punch (usually a shoulder tackle or flying elbow), or B for kick (usually drop kick). These will be the easiest and fastest moves to spam for the achievement. Running attacks against opponents on the ground, turnbuckle moves, jumping over the ropes to the outside, or slingshoting yourself over the rope into the ring all count as well, if you connect. Like many achievements in this game, only moves during your current play session are counted, so if you turn the game off, you'll have to start over.

I got this in my 7th match, without trying. just irish whip people in the turnbuckle and do a move, you don't need to climb up. when you get it the description is 'done 50 moves from or involving the turnbuckle' so there's no point jipping it.
