You'll need to complete a red charm. For that you'll need to get the requirement of that charm and insert the appropriate blue (level 2) charms, and for that you'll need to complete a few white (level 1) charms.

The requirements of the red charms are rather high; activate 20 wells is probably the easiest as you can play 3-1 over and over again, but it's up to luck when you'll get a red charm and which one.

I got one that required me to kill 600 named enemies, and didn't get another until I was well over that 600.

You have 2 spaces to equip charms. Once they are equipped, they can offer many beneficial effects, such as health regeneration and toxic pools that can damage/kill enemies.

Level 1: White
Level 2: Purple
Level 3: Red

To complete a level 1 charm, you must equip a charm by going to your equipment screen and pressing "back button" . Then press "LT" or "RT" to get to the charms tab. Cycle through the various charms you have available and press "A button" to select it. Finally, move it to one of the squares at the top.

To complete a level 1 charm, you need to do two things:

1. Complete the quest associated with that charm. This quest can be in the charms description and could be anything from kill X amount of enemies to kill X amount of leaders to activate X amount of wells and so on.
*Note: To quickly complete the charm quest for activating wells, load up the first part of the world "Serpent" (3-1), and activate the well. Return to the ship and repeat.
2. Under the quest description, there will be various rune shapes to insert runes. Press "X button" while the charm is highlighted and then press " A button" to insert them (providing you have that shape). The color of that rune shape (under the charm quest description) will turn from white to blue. Repeat for all slots on that charm.
*Note: The color and property of the rune don't matter so long as you insert into the charm. Once they have been inserted, they are gone for good and you get no benefit from the ruin, So try and use the less useful ones.

Once the quest is completed and all the ruins have been inserted, you will have completed your level 1 charm.

Level 2 charms are completed by completing the task for the charm, but you have to insert completed level 1 charms rather than runes into them.

Level 3 charms are the same as level 2 except you have to insert completed level 2 charms instead of level 1.

In addition to the other two guides that actually tell you how to complete the charm (which you should know by the time you get to Tier 3 charms), I'm gonna tell you the best way to get the charms and runes. OK, you're not gonna get a level 3 charm until you are a high level, already have some level 2 charms and have completed a fair number of checkpoints. Now, the best place to find charms are at the obelisks inside the NORN wells. After you've completed the game (or on a subsequent playthrough) if you go in to the NORN well in the World Tree in the AESIR there are around 20-30 of these obelisks in there which you will need all of the Cyberspace powers to access. Once I discovered this I managed to pick up 3 level 3 charms in 2 run throughs in there. Whether you get one that is easy to complete or not is a different matter. I've just completed one for 8,000 kills!

ENA - Find secret areas 15/15 - requires A, D and S lvl2 runes
ECotF - Kill any enemy 8000/8000 - requires S, A and R lvl2 runes
FoTHH - Achieve 200 hit combos 1/80 - requires a C shaped lvl2 rune
ECoTI - Earn all tokens for Hall of Heroes 0/3 - Requires R shaped lvl2 rune
SoA - Find secret areas 0/15 - no runes required
MI - Kill enemies with ballistic 2556/7000 - no runes required
ECotF - Kill goblins 40/10000 - requires S and symmetrical R lvl2 runes
SoA - Find secret areas 0/15 - requires C, S and symmetrical R lvl2 rune
ESAF - Destroy containers 164/300 - requires symmetrical R shaped lvl 2 rune
