How to unlock the Boss Rush Rank S achievement in Touhou Luna Nights - Definitive Guide
The Boss Rush will be unlocked upon defeating the final boss in the Extra Stage. However, you will most certainly not be a high enough level to achieve the S rank. (I was about level 26 when I finished the game)
To earn the S rank, you must complete the Boss Rush in 6 minutes. The final boss alone would take up all of that time at the average player's endgame level, so, that means it's time to get grinding. In the final hallway of the game, 2 screens left from the final boss, are a ton of powerful late game enemies. You will need to start from the right side where the healing vending machine and save point are, clear out this hallway with Thousand Dagger spell spam (remember, freezing time will make it consume TIME instead of MP), refill at the vending machine on the right side, causing the enemies to respawn, and repeat. There IS an EXP item available at the shop, but it is extremely expensive and really only gives you about 1 or 1 and a half worth of hallway clear-outs, and the jewels could come in handy later (read below)
I grinded to level 45 (only took about 1 hr 30 mins from level 26) and was able to complete the Boss Rush in 5min 51sec, so I would consider 45 the minimum requirement. If it's too tough, try grinding to 50 (or even higher!)
As for the Boss Rush itself, Thousand Dagger spam! You may have plenty of jewels from grinding, so sell all of those (I did however keep my Sapphires and Diamonds, as they boost Attack, which could help with the Boss Rush), and purchase TIME upgrades to hold more TIME. The first 3 bosses will absolutely melt under your Thousand Daggers and you will probably clear them out in well under a minute. Bosses 4 and up are where you will need to put in a bit more effort, but again, keep hailing them with Thousand Daggers. There isn't much more to say than that. If you fail to reach the S-Rank, you can always travel back to the endgame hallway and grind more levels.
And one last note. the final boss in the Boss Rush has a lengthy 5-7 second death animation, which DOES count on your clock. Be sure you are still in the 5min 40s range or low 5min 50s range upon defeating her.

Got it at lvl 41 doing 19 damage a knife, with 20 seconds to spare.

Just a question, why selling your jewel to get TIME ? I did the rush with constant spam of thousand dagger, and you barely hold time at all (since it doesn't stop the timer). I recommend keep the jewel instead, since mana regeneration and damage are much more important then time.
concerning the level, third try at level 46, you also level up pretty nicely during the bosses, so start practicing level 45 onward.

No need to grind. Thousand Daggers is so OP you can tank all the hits and heal off the graze.
I finished the entire game never selling a single gem.
Level 36-40 is all you need. Just rush every boss spamming Thousand Daggers and nobody stands a chance against you.
If you don't manage to beat the bosses fast enough you'll at least level up in the Boss rush and can.
Just get as close as you can to each boss and Spam daggers. If they attack while you're still throwing you'll just heal.
If you have bad luck and miss too many hits and lack enough MP just stop time and work on the MP recovery.
I started Boss Rush at level 32 and I'd gain 2 levels after each run but I was never killing them fast enough. At 36 I almost had it but the final boss froze me too many times to where I lost too much time. At level 38 I was strong enough to finish the Boss Rush at 5min and 45 seconds.

The only requirement for the S rank is to simply defeat all the bosses in under 6 minutes. You can use as many items as you wish and any abilities you want. My opinion is to get to level 40+ before attempting this to make short work of the bosses. I would use my farming method of using the holy sword and go right of nitoris shop and kill the 6 blue orbs and go left back into the shop, and repeat until high enough level. Once your strong enough, all you need to do is stay stocked on all health potions and max out your time and daggers. Then start boss rush from the shop menu and spam thousand knives! Remember thousand knives does less damage during time stop, so spam a couple in normal time, then a couple in stopped time, get the hang of mixing it up. I found a video Ill link here that shows someone beating in over 5 minutes to get an idea of the strategy.
