If you can't win sprint stages, doing this on a climbing stage with your best climber is fairly easy. If there are two mountains within the last 60 kilometers, simply attack up the first of the two mountains at 100% effort, and you should be able to keep the lead untill the end. The best climber in the game is Contador so this is probably the easiest with him.
