Chapter 4 includes Samples 16 - 20 and Chapter 5 is samples 21 - 25. To qualify for an "Extra Reaction" you need 5 total chain reactions. A chain reaction occurs when 3 colours disappear and the colours that come together form another group of 3 or more of the same.
I found this was easiest by making sure the Puzzle setting is on easy to reduce the colours in the chain and then prepping the chain starting form the entry point to give you time to add more colours before starting the chain. Although this could happen randomly that is unlikely.
An Extra Reaction popup will occur when the 5th chain reaction happens. I completed it on Sample 4 of Chapter 1 and will update my thoughts on the best level to do this in on Chapter 4/5 when I complete that.

Edit: *This should be fairly doable in all levels but I found it was easiest to stick with levels that have slower moving puzzles. I completed it on Sample 16.*
