Fight against every character over Xbox Live (Player or ranked). Win or lose, doesn't matter, you just need to finish the match. If you want to boost, get a friend to help you by picking every character in order until you've fought them all. You do the same so the opponent gets the achievement as well.
NOTE: DLC characters don't matter, not required for the achievement.

Good job on the "NOTE" because I was worried Jill & that Green tenticles dude (forgot his name) would be required since the person I was boosting this w/ in player match does not have them unlocked.
So I was able to get it w/o him having them to choose from on his roster.
