To unlock Randomazzo enter the Gallo Tower level and head north. You should see an icon on the map that looks like a few playing cards. Pick that up and you should be all set.
n.b. Gallo Tower is unlocked by levelling any character to 60 in the Dairy Plant level.

This is located in Gallo Tower, Stage 4. Simply follow the green arrow up all the way to the relic, whose icon looks just like in the achievement image. Grab it and this achievement will unlock after the run is over.
