The player can unlock Sir Ambrojoe by killing a total of 6000 Stage Killer and Elite Stage Killer (per save, cumulative over multiple runs), then purchasing with Sprite, which scales with other characters purchased.
Alternatively, Ambrojoe can be unlocked in the Forbidden Scrolls of Morbane by casting the Spell "languorino".

There are five secret characters that each require you to reach level 80 with them. This can be done on any stage, and in my opinion the easiest one is Inlaid Library, Stage 2. Each of these five characters has a unique starting weapon, one that cannot be evolved.
If you want to ensure you reach level 80 with each character, you should unlock and enable Endless Mode just to be safe. Refer to Greatest Jubilee (50G) for unlocking Endless Mode. Enable Endless Mode on whichever stage you play just to be safe, so you don't have to worry about Red Death ending your run before you get to level 80.
By this point, you likely already have a build you like to use that'll get you to level 80 without issue, but once again, here is the one I recommend:

- Starting weapon (not possible to evolve)
- Spinach
