The Moongolow bonus level is a reward for unlocking hyper mode for 4 levels. Hyper mode is unlocked for a level once you beat the boss that spawns at 25 minutes. Meaning to do this, you need this achievement:
You have to play Moongolow more than once to survive for 15+ minutes.
The first time you play Moongolow, just as the clock is hitting 15 minutes, the level will go into a special sequence, and you'll be teleported into a new area without any of your upgrades. You must run through this passageway to get an item at the end (the Yellow Sign).
If you die during the running part, you do not have to replay Moongolow in its entirety and can select this sequence on its own, called Holy Forbidden, from level select. Holy Forbidden will disappear from level select once you complete it.
Moongolow will continue to teleport you to Holy Forbidden at 15 minutes until you complete this sequence and get the Yellow Sign, earning this achievement:
Once you get the Yellow Sign, you can replay Moongolow, where the level will no longer end at 15 minutes.

Very well explained. I played the level a couple of times before reading this guide, and I will be honest, I had no idea what was going on or if the corridor ever ended etc.. after reading the guide it was easy to get. Thanks.

Each of the Skip achievements is for surviving until the "end" of a bonus stage. Refer to Boss Rash (5G) for unlocking all of the bonus stages.
The first three bonus stages (Molise, Moongolow, and Green Acres) don't have anything special about them that impacts how you play them in any way. The fourth (Bone Zone) and fifth (Boss Rush) are a bit different, and are covered in their own achievement solutions.
Since these aren't really much different from normal levels, all the same strategies apply for surviving until the end of the bonus stage: get your three best evolved weapons (Santa Water, King Bible, and Axes), as well as the evolved Lightning Ring, Spinach, and whatever your starting weapon is, and you shouldn't have any issues surviving until the end of each of the first three bonus stages.
Note: The Yellow Sign relic is obtained in Moongolow by reaching the end of the stage while playing as a normal character. It's therefore most efficient to do your 15-minute run of Moongolow while playing as a non-secret character. My recommendation is Pugnala, as she's fast, strong, and has good starting weapons.
