The Train is an interactable specific to the map Laborratory.
First load up the Laborratory map, See how to unlock it here:
Then run along the bottom of the map, activating the levers along the track as you go. This will cause the train to rush through and damage enemies. The amount of damage dealt is difficult to track, so you wont know when you hit 25120 but a dozen or so activations should do it. This does appear to be cumulative too so can be done across runs.
Once you end the level the achievement should unlock if the criteria is met.

Along the bottom of this map is train tracks which you can activate by touching any of the levers along the track. The levers reset too, so feel free to just bounce back and forth between two levers.
There's no way to track how much damage you've done with the train, but this is cumulative across all runs. Personally, I used it about a dozen times and unlocked this achievement in one run.
