This achievement is story related, and it is unlocked by defeating Mordra the Broodmother, which is the boss of The Den of the Broodmother. It's located at the top left of the Royal Gardens (when looking at the map). It wasn't accessible until I was level 16; although, I'm not sure if that was because I was playing on hard.
You will know you're at the boss when you get to a ledge that is overlooking a bunch of spider eggs. The fight starts once you jump down. All I did was have a shotgun (The Repeater) equipped, and I ran in a big circle while focusing on the smaller spiders first, which was quite easy considering the actual boss moves pretty slow. After the smaller enemies were destroyed, I just kept going in a circle around the boss while shooting it and occasionally hitting it with my overdrive.
