How to unlock the Angel of Death achievement in Warframe - Definitive Guide
Figured I'd post a little something even though this achievement is pretty simple.
Best way that I've found to get this early on is on the Earth> Pacific mission.
Easiest done solo. *Or if you have a friend or group of friends who will not go for kills, it can also be done. Plus then they can also revive you if downed.
Just (As in any mission if you want more kills) let the enemy activate alarms so that they come looking for you.
There's no time limit so take as long as you need until hitting 100.
The achievement will unlock as soon as the "100 Kills in a Single Mission" Challenge update pops up.
At the end of the mission I had 111, and it was probably still possible to go further.
Hope this helps anyone who's wondering about it.
**After some kills(Maybe 10-20) the area you are in will deplete of enemies. Don't fret, just move to another part of the map, without disabling the alarm. That area will have more enemies, and after that area you will have to move again, either a new room or the previous one because enemies should be respawning depending on how long you took.

I actually didn't have to use any. Just my bow.
For newer player, possibly could be required. But I also have a good amount of time played on PC so I have a little bit of an advantage I guess.

I actually did this on the very first Earth mission after the tutorial.
The mission entails holding a position, and the enemy seems to have an infinite spawn if you let them take it and, instead, pick them off at distance.
All I did was hang back and use the bow (awesome weapon BTW) as I was trying to get use to the game, and I had this done before the mission ended.
Questions, corrections and neg vote justifications below.

Name of the mission would be helpful.

Sorry bud - I do not remember the name of it. It was a 1-3 mission directly after the tutorial - that is all I remember.
HOWEVER, I have found that nearly any mission that is not an "Exterminate" type will have a constant flow of enemies - if you solo a capture mission, a sabotage mission, or basically anything that is not "Exterminate", and take the time running around the map, you should find 100 fairly easily.

The first planet (Mercury) has a Horde-style Lares mission (Defense). Start it solo, and by the sixth chapter you will have shot a hundred. It is not necessary to complete the mission. Level must be at least 7, but higher is better.

Sooner or later you will make this trophy on defense or survival.

Got it in the Mission to collect the Forge, there are a lot of enemies running around, let them call for reinforcements and there are more mobs, before evacuation you need to kill 37 enemies, you only have to find 63 in the mission

I did some clearing on the ground on the second mission, just stood between two houses of enemy kubrows, and mauled them with melee weapons until the trophy fell out.
