There's a vortex grenade just before the boss room, fall straight down at the start of the long corridor where the walls close in. Throw that at his feet to take off a massive chunk of health straight away.

There are multiple of these throughout the campaign. When they appear you have to beat them to progress, so you can't miss them. First appears towards the middle of chapter 2.

With the DLC Update, the Great Unclean One can also be found in Horde Mode. Every 3 rounds in Horde Mode there is a boss encounter. For the Great Unclean One, I encountered him during Wave 6.
