I found the easiest mob to get this from quickly was the Demonhost. The Demonhost is a monstrosity that doesn't spawn on every map so it may take you a few missions until you find one.
The higher the difficulty the more damage the Demonhost will do
Best to do it with the Ogryn with a shield or Zealot with a lot of health and/or stamina. Just aggro the Demonhost and hold your block button whilst using a melee weapon.

I'm pretty sure there's more to it than just the way it's worded here. Tried with the Ogryn 3 times and no achievement so I'm thinking it either has to be done with some specific curio equipped or do it at a specific level/difficulty level

I just unlocked it earlier with ogyrn against a horde I was in a heresy match but I think it's more so the higher the difficulty the more damage they deal to you

The demon host will definitely make this trivial (including the 900 damage Penance above this one). As another, more accessible, option though. Simply go on a damnation run and find a large melee group and block. You may not get it just by holding block, especially if you have lower stamina, but there's more though enough tim,e to let some Regen and block again.
