Throughout the DLC you'll encounter Explosive Barrels, characterised by red lights on them.
While there are more than 50 barrels, to be safe make sure you shoot them at every opportunity to ensure you have enough before you complete the DLC.
If you shoot every one you encounter then you should get this achievement sometime during Act 2.

Just worth noting that this is cumulative across all playthroughs.
Managed all achievements except for this one during my first playthrough. Went into the DLC section of the main menu and started a 2nd playthrough and got it on my 6th barrel. First 5 where you pick up the Ray Gun and makes you fire at the barrels then unlocked it on the next one I shot right after them so don't worry, you don't have to get 50 all in one playthrough.

When you first receive the Ray Gun, there will be a quick tutorial about blowing up yellow Explosive Barrels. Keep blowing up more throughout all three Acts to unlock the achievement.
