I earned the second rune in Chapter 2, in the Bayou level. I defeated an outlaw and the rune dropped.
It said it was a "Power Rune" and would make getting around a little easier. I believe it will help break the metal barrier found in the level the Hunt (though I have not confirmed this, and others have said that is sometimes bugged). I'm not certain if it's obtainable in every run to the Bayou in Chapter 2 until you earn it or if it is random.
