Loot supplies are the ammo, armor and silver coin drops from dead enemies. Just collect 250 of them by simply walking over them. Got this allready after the third mission. No need to boost or something.

This is cumulative and will unlock naturally while you are playing the game as you will pick up more than 250 while playing through the campaign. This includes ammo, health, coins, armor and weapons. If they are placed in the environment, you need to press 
to pick it up, but if enemies drop anything, simply walk over them to pick them up.
This is not tracked separately in the ‘Statistics’ menu.
