This can be one while doing your 180 Rivalry wins. As you are doing those it will count towards this so don't bother with regular matches and just play the rivalry ones.

This can be done at the same time as "Everybody wins!" and "Match maker" to save yourself some time. You simply have to play 50 matches from the card in WWE Universe Mode. You can change the match and participants by pressing 
if you are also going for "Everybody Wins" and "Match Maker".
Please note that this can only be done single player and using a second controller will not count towards your win total (I found this out the hard way!)
