This must be done against an AI controlled opponent and can be done on any difficulty. Go into a One on One match and choose anyone. Play the match as normal until you get their body parts red. Holding the A button will initiate a submission move grapple. Mash A when the minigame pops up on the screen. If the AI's body damage isn't red, it's pretty much impossible to make them submit. Each character has different submission moves that damage different body parts, so try out the submissions at the beginning of the match to see which body parts they damage if you don't already know. You can do submissions to either a standing or grounded opponent. Just make sure you win with a regular submission and NOT a Signature or Finisher.
