As described in the description, 50 enemies must be killed. It does not matter which mode you are in or whether a team member has already inflicted damage.

The trophy will pop upon your fiftieth kill which will likely be earned within 5-10 games depending on how well you perform. The trophy will likely be one of the first trophies you earn and will come from just playing the game naturally.
