To unlock this Mode, simply beat the Game once.
You can use cheats to make this
Achievement.
On Main Menu, highlight the Options, and make the sequence:
LT + Left + Y + Start.
And activate Infinite Health, Ammo and Grenades.
The Achievements pops when you reach a 800k ~ 1M of Score.

I had already tried this and quit at 953k without getting it. On my third attempt, I got it at 937k. I think the enemies spawned vary, thus the score for 25k enemies will vary.
I actually died on my second attempt, despite using the infinite health cheat. The red version of the enemy resembling a xenomorph (from the Alien film series) leaves a puddle of goo after death. Contact with those puddles reduced my health while using infinite health. They're the first enemies you'll meet in the Nest, and the puddles hurt there too. If anyone can confirm this, it may be worth noting in the solution to save someone a dull replay.
