I played the game on hard first, so I am not positive if there are any characters not unlocked differently during a playthrough of the story on normal first.
All but two characters unlock as the story progresses. You unlock Beast Boy for completing normal and Rocket for completing hard.

Nope, I did the game on hard first and only Rocket unlocked. Everyone else seemed to unlock during the story, but like I said, I did hard first, so not sure if some others are hard only compared to normal.
