A signature creature is the one in the picture next to the opponent before the battle. When your opponent plays this card, a little animation will happen where the card picture will enlarge.

The picture changes for everyone except the people at the top of each ladder tree and the achievement only seems to unlock against those at the top so I will list the cards so you can check to see if you have the one of them already.

I assume that this can also be done in Expert Mode.

You have two options:
1. Have the Card in your Deck and attack the opponent with it
2. Use an effect to take control of the creature and attack the opponent with their own card. You can use cards such as Creature Seizure, Creature Swap, Brain Control or Autonomous Action Unit

I will sort the opponents by series.

Series 1: Yu-Gi-Oh!
Ladder 1: Maximillion Pegasus has a Relinquished. Relinquished needs to be ritual summoned using a card such as the one in the link or Advanced Ritual Art.
Ladder 2: Seto Kaiba has a Blue Eyes White Dragon.
Ladder 3 Marik Ishtar has a Legendary Fiend.
Ladder 3: Yami Yugi has a Dark Magician.

Series 2: Yu-Gi-Oh! GX
Ladder 1: Sartoruis Kumar has an Arcana Force EX - The Light Ruler.
Ladder 2: Yubel has a Yubel - The Ultimate Nighmare.
Ladder 3: Nightshroud has a Red Eyes Darkness Dragon
Ladder 4: Jaden Yuki has an Elemental HERO Neos.

Series 3: Yu-Gi-Oh! 5D's
Ladder 1: Jack Atlas has a Red Dragon Archfiend.Note that this is the Synchro version, not the XYZ version. I have no knowledge if the XYZ version would work.
Ladder 2: Aporia has a Meklord Astro Dragon Asterisk.
Ladder 3: Zone has a Sephylon, the Ultimate Timelord.
Ladder 4: Yusei Fudo has a Stardust Dragon. Note that this is the Synchro version, not the XYZ version. I have no knowledge if the XYZ version would work.

Series 4: Yu-Gi-Oh! ZEXAL
Ladder 1: Kite Tenjo has a Galaxy-Eyes Photon Dragon.
Ladder 2: Astral has a Number C39: Utopia Ray.
Ladder 3: Vector has a Number 104: Masquerade.
Ladder 4: Yuma Tsukumo has a Number 39: Utopia.

I'm glad you posted that you have to attack. I summoned teo blue eyes against kaiba and no achievement. I couldn't attack as i was using gravity bind to allow room to summon the Blue eyes. He ran out of cards and i won. Next duel i got blue eyes on turn 3 attackedand then chievo popped!!! Great guide :)

Honestly, I am no expert at Yu Gi Oh. I only know the first two series. I don't think you need to buy any DLC as you win decent cards.
I did create a burn duck to help with expert mode

I got this by playing Yami Yuji a second time and taking control of his specialty creature the Dark Magician and using it against him. You would need a card that takes control of an opponents monster, I used "Creature Seizure" which allows me to choose a normal monster of mine to switch with the normal monster of the opponent. Each character only has one specialty creature, its all in the heart of the cards from there. Happy Dueling !

Edit: Having a signature creature belonging to the duelist in your own deck and using it against does work also, thanks S2N Diabolical for clearing that up.

Do we know if you could just have the opponents signature creature in your deck at the start of the duel?

That's the thing I was going to try but getting a signature creature from the duelists are hard unless you have one from deck in DD or got lucky. I got Summoned Skull from him but that wasn't even considered a signature. Lemme know if you get it and ill edit the solution thanks.

Nope its called Creature Seizure haha, I never heard of it either but its there and it only works if switching between two normal monsters.

Yes having their signature creature in your deck counts. I attacked Seto Kaiba with my own Blue Eyes White Dragon and got the achievement.

Haha I personally wished I had gotten a brain control but whatever works, that was the quickest way for me since its really random to get a specialty card unless you already had it .

Confirmed. Using Gaia The Fierce knight against Solomon Muto won't unlock the achievement.

your best bet is to stick with the people at the top of the ladder. the people along the way don't seem to count. I tried multiple early ppl on a ladder on the first day of launch. Luckily both Yami Yugi + Dark Magician or Seto Kaiba + Blue Eyes White Dragon are fairly "simple"

I used Autonomous Action Unit to steal Dark Magician from Yami Yugi's graveyard and attacked him with it.

Is the card pictured next to them their signature creature or is it something else? I want to beat duke using Graceful Dice but don't want to waste a game trying to get it since its not a creature and not sure if its his signature.

I also just got this achievement from yami yugi I use a spell card called creature swap
for his Dark Magician

Each player chooses 1 monster they control and switches control of these monsters with each
