Start a game on Easy and make it to the boss room, after defeating the boss DO NOT enter the portal to the next floor, instead find some lava or spikes and walk back and forth until you die.
Since some boss rooms don't have hazards like this, you are also free to go to any other room on the map to die. (re: UnsungGhost)
You can also kill yourself with Grenades if you are on really low health.
