How to unlock the Headshot! achievement in ZOMBI - Definitive Guide

You obtain the gun as soon as you get the BOB so this can be obtained more or less the first time you encounter a zombie after that. Should be the first combat achievement you receive.

Everything TheGearGuardian stated is true. I just want to give a quick note on doing this easy if you have any issues under pressure. After you get back from getting the backpack you have a second mission. Once your friend on the com helps with the sliding verticle doors .... You can see a mounted gun. If you interact with it he will advise that you have inbound zombies. If you head back under the door(behind you that you came through), the zombie will get on the ground to crawl under after you. This makes for easy aiming and achievement w/o trying to react quickly.

I just used the mounted gun to headshot a zombie and the ach unlocked.

You are given a gun early on in the game once you obtain the BOB and the Cricket Bat. In order to earn this trophy, shoot a zombie in the head with a gun and the trophy will unlock. Bear in mind that the zombie does not need to die just as long as you shoot it in the head once.
